Measure the clear space between the posts (inside face to inside face) along the rail, and the width of one baluster as you look along the railing.

Equal gaps gives the fewest balusters that keep every space at or under your max gap, with all gaps exactly the same. Cut a spacer block to the gap length and use it between each baluster, or hook your tape on the post and mark each position.

Set on-centre keeps a spacing you choose and centres the layout, so the gaps at both posts match.

For a stair railing, pick Stair and enter one step's rise and run. Balusters stand plumb, so gaps are worked out level (the way the code measures openings) and the marks are given along the rail.
